the intiatl number order is 150 jets and it almost certain that it will go up to 250.

whether more order will be placed beyond 250 depends upon maturity of any 5th generation fighter for PAF and its induction timeline.
Also the FC-20 final number currently being quoted as 72 being planned, will play a vital role.
if FC-20 numbers go up, i don't see any increase in JFT beyond 250.

as for second part of your question, determinism a GOOD number to secure regional/national interest is something that cannot be determined. all lot of factors come into play, from first and foremost the government policy toward any destabilization attempt to the details of adversary!!

How many fighter 4th gen fighter jets would we need if we were to become a key regional power considering India is equipping 4th and 5th gen jets? Keep the question of resources/funding/etc out for now, just consider ideal projection.

Secondly what leaverage FC-20 induction would provide to the PAF considering weapon package similarities with JF-17 program, both being single seat jets et al.
 
to be a regional power , there are a lot of factors to be taken into consideration other than economics.. radius of potential influence, location of our country [to attack and to defend] , number of days we can maintain in war [both offensively and defensively] , technological/training edge , resolve , logistics , key allies , ability to sync different military fields effectively ...... to name a few-- unfortunately economics and politics are not the strong suit of this government and have done longterm damage to pakistan's interests---

airforce is just a small part of the puzzle , however , its very important as , as opposed to the navy , the airforce can be upgraded to a very potent /state of the art force at a fraction of time and money-- however the army/navy provide the physical presence in the region to maintain / project power-- so a balance is needed amony the forces depending on political aspirations and geography of a country-- pakistan has a small coastline and no power projection aspirations , so the easiest way to go is to have missiles and good fighters to complement the other forces

the 21 squadrons / 400 fighters that paf is sanctioned to operate are a good number -- will make paf even more capable e long range precision weapons aswell has 4+ fighters that will hopefully replace all the old fighters

the design [delta + midmounted canard] of fc20 gives certain aerodynamic capabilties which are not so pronounced in the jft design and viceversa --- coupled with the fact that fc20 can hold bigger/more capable radar, more fuel/more payload to independently operate at longer distances, thus touching more iaf airbases than as of today-- will give another dimention to paf to exploit
