Honestly I don't see Karachi being province. Its case is similar to Mumbai. Marathis will never allow Mumbai to become separate state. They even have quota in private sector there.

Keeping in mind to break the provinces on geographical and economic grounds not ethnic ones.

Divide solely on administrative terms. No ethnicity, no caste, no region bullshit. We are Pakistanis and Muslims and those who say otherwise can go and screw their screws...
Divide whatever makes it easy for people to enjoy better access to governance.
